Post : Scientist - New Drug Target Validation

Position Summary
Known for its scientific and operational excellence, Regeneron is a leading science-based biopharmaceutical company that discovers, invents, develops, manufactures, and commercializes medicines for the treatment of serious medical conditions. Regeneron commercializes medicines for eye diseases, high LDL-cholesterol, atopic dermatitis and a rare inflammatory condition and has product candidates in development in other areas of high unmet medical need, including rheumatoid arthritis, asthma, pain, cancer and infectious diseases.

Summary
This position in the Pre-Therapeutic Target Discovery (PTTD) group will be responsible for conducting preclinical studies to validate and advance potential new therapeutic target proteins. This scientist will utilize basic research methods, including DNA, RNA and protein expression assays, histology, 2D and 3D imaging, and flow cytometry, applied to various transgenic and conditional mouse models, cell lines, and human tissue specimens. A background working with genetically modified mouse models, and a solid general knowledge of developmental biology, functional anatomy, and immunology, is highly desirable. The candidate will be responsible for leading studies, effectively interpreting and communicating data, and collaboratively reaching conclusions about the therapeutic potential of targeting proteins with antibodies or other drugs.

Responsibilities
(i) Develop genetically manipulated mouse models—such as null, conditional null, transgenic, mutant, oncogenic—for assessing secreted and membrane-bound proteins as potential therapeutic antibody targets
(ii) Perform in vivo and ex vivo assays on these mice and their tissues, perform analyses on the results, and formulate conclusions on the suitability of specific proteins as antibody targets
(iii) Utilize these model systems to provide a better understanding of basic biology, and the pathogenesis of disease.
(iv) Participate in a screening process to evaluate the phenotypic characteristics of mice with targeted deletion of individual genes as a way to identify potential new targets.
(v) Develop new methods to improve and extend our mouse phenotype evaluation capabilities, including methods to conditionally delete genes in specific tissues and/or at specific stages of development
(vi) Evaluate newly generated antibodies for sensitivity and specificity in detecting targeted proteins by histology, Western blots, flow cytometry
(vii) Maintain meticulous records and prepare data for presentations, both internally and at external scientific conferences
(viii) Write documents to support patent applications, external conference posters, and journal articles

Candidate Profile
This position requires a minimum of a PhD or MD with a background in mammalian systems biology, and 2-5 years of relevant postdoctoral experience. Additional requirements include:
• Ability to design well-controlled studies and to apply the highest scientific standards in conducting those studies and evaluating their results
• Proficiency in at least some of the following lab skills: animal handling, drug delivery, tissue and/or embryo harvest, histology, digital image acquisition, DNA, RNA and protein expression assays, flow cytometry and FACS, cell culture
• Specific training and experience in generating and analyzing genetically modified mice (transgenics, knockouts, LacZ/GFP reporters, Cre-deleters) is desirable
• As this position involves thorough investigations of mouse models, broad knowledge of a variety of biological systems is a plus
• Good team skills: communication, collaboration, supervision of research associates
• An appreciation of the need to apply rigorous analytical methods is essential
• Ability to present clear, well organized project updates to supervisors and other group members, as well as to colleagues of diverse science backgrounds in other departments
