Animated Video Representation: HPLC (High Performance Liquid Chromatography): Working and Instrumentation

Clinical courses

High-performance liquid chromatography is a technique in analytical chemistry used to separate, identify and quantify each component in a mixture.
The method relies on pumps to pass a high pressure liquid solvent containing the sample mixture through a column filled with a solid adsorbent material.
Affinity of each component in the sample is different with the stationary phase or adsorbent material, causing different flow rates for the components and due to this differences separation of the components takes place as they flow out the column.

In this animated video various parts of the HPLC instrument is discussed which are:
1. Solvent Reservoir
2. HPLC Pump
3. Vacuum Pump
4. Degasser
5. Solvent Mixing Valve
6. Pre-column (Guard Column)
7. Sample Injector
8. Analytical Column
9. Detector
10. Recorder
Working of HPLC instrument is explained by animated visuals.
