The Dr. Reddy's Institute of Life Sciences was conceived in the context of globalization, to foster research competence in India and to bridge the divide between academic research and the pharmaceutical & biotechnology industries.  The institute was incorporated as a non-profit entity under Section 25 of the Indian companies act, facilitating operational autonomy. Construction of the DRILS building comprising 44 labs and an administrative block and spanning ~130,000 sq.ft. in area, began in 2004 and was completed in January 2007.

Project Title: Use of DNAzymes to improve CRISPR-mediated knock-in.

Candidates with a keen interest in targeted gene modification and prior hands-on experience with molecular biology and cell culture are preferred. Experience with CRISPR would be an additional advantage. A possibility of Ph.D. enrolment exists, but is not guaranteed. Work will involve DNAzyme construction, DNAzyme library creation and verification, recombinant protein purification, CRISPR reagent construction including gRNA design, in vitro transcription, and extensive mammalian cell culture, transfection, and screening including analysis of viability and marker expression.

Qualifications:
• Graduate Degree in Professional Course with NET qualification
• Post Graduate Degree in Basic Science with NET qualification
• Post Graduate Degree in Professional Course Acceptable NETs: CSIR-NET including LS; GATE, JAM, GPAT, BET, JEST, JGEEBILS, ICMR-JRF and ICAR- AICE.

Emoluments and Duration : Rs. 25000+HRA per month

Project duration: Three years. Appointment will be renewed yearly based on performance

Project Summary
CRISPR-Cas9 technology is a recent advancement in genetic engineering allowing targeted gene disruption with high efficiency. The technology can also be used for targeted insertion of DNA sequences, but less efficiently.
The overall goal of this proposal is to use DNAzymes to facilitate high-efficiency targeted gene insertion. Specific objectives:
i. Proof-of-concept studies using a DNAzyme targeting DNA ligase IV, to inhibit NHEJ and promoter HDR mediated gene insertion.
ii. Construction of the randomized DNAzyme library in a vector suitable for in vivo expression in cell lines.
iii. Screening and identification of HDR enhancers
iv. Validation of the top 5 hits from the screen for further characterization and testing.
